MSTS activity editor works as part of the main program like the one in TSRE5, so you will need all of the MSTS base files, excluding routes, consists and trainset in order to run the editor.

I've used TSRE5 to build a number of activities...I have built Amtrak's Southwest Chief run from Chicago to Los Angeles over 10 different routes. Some of those activities will not run under MSTS because I used some features that only appear in OR.. But Yes, I've used it...
