The larger tank contained the spray, the other was an air tank fed by the locomotive air brake line. This charged air propelled the weed-killer liquid into nozzles fed through plumbing under the car, and sprayed along the right-of-way as the train rolled along. Typical consist would be a locomotive, the weed sprayer, and a caboose for the crew and to provide rear-end protection.
